If I have many files already in my media library, all of which are already used in at least one post, page, forum thread, profile page avatar, etc. Does moving files around different folders affect all of the posts and pages that the media is already used on? i.e. Will I have to go to each post and manually re-add each media file from the new folder?

Pups sorry I missed the first post notification.
Put existing images that are called in content won't change anything to your content. To make virtual folders we use a WP taxonomy so links to images remain untouched
